High-throughput screening for modulators of cellular contractile force.

چکیده
When cellular contractile forces are central to pathophysiology, these forces comprise a logical target of therapy. Nevertheless, existing high-throughput screens are limited to upstream signalling intermediates with poorly defined relationships to such a physiological endpoint. Using cellular force as the target, here we report a new screening technology and demonstrate its applications using human airway smooth muscle cells in the context of asthma and Schlemm's canal endothelial cells in the context of glaucoma. This approach identified several drug candidates for both asthma and glaucoma. We attained rates of 1000 compounds per screening day, thus establishing a force-based cellular platform for high-throughput drug discovery.

Nootropic Medicinal Plants; Evaluating Potent Formulation By Novelestic High throughput Pharmacological Screening (HTPS) Method
The principle of this method was to screen the pharmacological activity of six prepared polyphyto formulations by using high throughput screening method for their nootropic action. The study was performed in three stages using one, two and three animals, respectively in a group. Test formulations were given p.o daily at the dose of 50 and 100 mg/kg body weight. The test formulations were compar...
متن کاملCellular Assays for High-Throughput Screening for Modulators of Trk Receptor Tyrosine Kinases
Trk receptor tyrosine kinases are required for signal transduction initiated by neurotrophins leading to cell proliferation, differentiation, survival and death. Alterations in Trk kinase activity have been linked to various diseases. To address the need for cell-based assays for screening and studying the selectivity of Trk kinase modulators, we developed high-throughput cell-based assays for ...
متن کاملScreening for Small-Molecule Modulators of Long Noncoding RNA-Protein Interactions Using AlphaScreen
Long non-protein coding RNAs (lncRNAs) are an important class of molecules that help orchestrate key cellular events. Although their functional roles in cells are not well understood, thousands of lncRNAs and a number of possible mechanisms by which they act have been reported. LncRNAs can exert their regulatory function in cells by interacting with epigenetic enzymes. In this study, we develop...
متن کاملTherapeutic Discovery A Robust High-Content Imaging Approach for Probing the Mechanism of Action and Phenotypic Outcomes of Cell-Cycle Modulators
High-content screening is increasingly used to elucidate changes in cellular biology arising from treatment with small molecules and biological probes. We describe a cell classifier for automated analysis of multiparametric data from immunofluorescence microscopy and characterize the phenotypes of 41 cell-cycle modulators, including several protein kinase inhibitors in preclinical and clinical ...
متن کاملUse of FLIPR Membrane Potential Dyes for Validation of High-Throughput Screening with the FLIPR and μARCS Technologies: Identification of Ion Channel Modulators Acting on the GABAA Receptor
Fluorometric imaging plate reader (FLIPR) membrane potential dyes (FMP-Red-Dye and FMP-Blue-Dye) were evaluated for the detection of compounds acting either as positive allosteric modulators or agonists on the GABAA receptor (GABAAR). A stable HEK293 cell line with constitutive expression of the rat GABAAR α1, β2, and γ2 genes was used to establish a functional high-throughput screening (HTS) a...
